P51. Identification of prostate cancer-associated antigens by oxygen manipulation
نویسندگان
چکیده
Background Therapeutic vaccination against prostate cancer (CaP) remains marginally effective. A reason for failure may stem from the fact that vaccine cells are usually cultured in ordinary air. Solid tumours including CaP contain regions with oxygen deficiency (hypoxia) secondary to the lack of blood supply to the growing tumour nodules, which may lead to changes in expression of cancer-associated antigens in tumour cells. To test this hypothesis we determined whether oxygen-sensitive CaP-associated antigens in cultured CaP cell lines and human tumour tissues exist.

Novel identification of the ETS-1 splice variants p42 and p27 in prostate cancer cell lines.
ETS-1 is involved in cellular functions such as proliferation, migration, invasion, apoptosis and angiogenesis. The ETS-1 gene encodes three distinct proteins, ETS-1 p51 encoded by a full-length mRNA, ETS-1 p42 and ETS-1 p27 encoded by an alternatively spliced mRNA lacking exon VII and exons III-VI, respectively. ETS-1 p51, commonly considered to be the active form, has been studied in prostate...
متن کاملIdentification of Intracellular Sources Responsible for Endogenous Reactive Oxygen Species Formation
The endogenous reactive oxygen species ("ROS") formation is associated with many pathologic states such as inflammatory diseases, neurodegenerative diseases, brain and heart ischemic injuries, cancer, and aging. The purpose of this study was to investigate the endogenous sources for "ROS" formation in intact isolated rat hepatocytes, in particular, peroxisomal oxidases, monoamine oxidase, xanth...

متن کاملIsolation of human prostate cancer cell reactive antibodies using phage display technology.
Here we describe a phage display strategy for the selection of rabbit monoclonal antibodies that recognize cell surface tumor-associated antigens expressed in prostate cancer. Two immune rabbit/human chimeric Fab libraries were displayed on phage and used to search for tumor-associated antigens by panning on DU145 human prostate cancer cells. For this, we developed a novel whole-cell panning pr...
